Characterization, expression analysis and RNAi-mediated knockdown of two aquaporin genes in the cotton leafworm, Spodoptera littoralis (Lepidoptera: Noctuidae)

Aquaporins (AQPs) are integral membrane proteins that can transport water and small molecules across cell membranes in organisms. In a previous report, we identified an AQP (SlAQP1) gene of the cotton leafworm, Spodoptera littoralis. current study, identify characterize two more SlAQP genes their developmental tissues expression. Predicted amino acid sequence phylogenetic analysis revealed SlAQP2 is glycerol channel belonging to insect specific Eglp subfamily, whereas SlAQP3 Prip subfamily. expression was detected mainly first four larval instars adult males, all stages. midgut Malpighian tubules male testes, tested. RNAi mediated knockdown each separately resulted deleterious effects including pupal mortality, deformed pupae adults prolonged development. Results were confirmed using qRT-PCR, which downregulation both after injection larvae with dsRNA. Our results confirm presence AQPs S. littoralis suggest has role reproductive ability.
